Effects of allopurinol on the survivals of paraquat intoxicated rats and cultured hepatocytes

Abstract
We Investigated the effects of allopurinol, xanthine oxidase inhibitor on paraquat toxicities in rats and cultured rat hepatocytes. The survivals of paraquat-intoxicated rats were increased by allopurinol treatment compared to paraquat-treated control rats. Xanthine oxidase activity in the liver of allopurinol-treated rats was decreased significantly as compared to normal control rats(P£¼0.01). In primary cultured hepatocytes, the viability of paraquat-treated cells was significantly Increased by allopurinol treatment ( P£¼0.01). Those results suggest that paraquat-induced toxicity can be reduced by administration of allopurinol, indicating that this toxicity may be associated with the formation of paraquat radicals by xanthine oxidase. In this study, we obtained the results of the in-vivo paraquat toxicities were similar to that of the invltro paraquat toxicities. So we can study the paraquat toxicities on the in vitro cultured rat hepatocytes in detail.
